Identification of transcripts by macroarrays, RT-PCR and in situ hybridization in human ejaculate spermatozoa.

چکیده
Round spermatids contain high levels of extremely varied mRNAs that are synthesized either throughout early spermatogenesis or during spermiogenesis from the haploid genome. Concomitantly, with major changes in the chromatin organization, arrest of transcription occurs at midspermiogenesis. However, previous investigations using RT-PCR have revealed the persistence of numerous and different transcripts in ejaculated spermatozoa. In the present study, a step-by-step analysis by means of macroarray hybridization, RT-PCR and in situ hybridization was performed to identify more accurately the different mRNA species found in the human ejaculated spermatozoa. The data showed an extended pattern of various transcripts encoding a diverse range of proteins involved in signal transduction and cell proliferation. For the first time, they demonstrated that mRNAs coding for the transcription factors NFkappaB, HOX2A, ICSBP, protein kinase JNK2, growth factor HBEGF and receptors RXRbeta and ErbB3 accumulate within the sperm nucleus. The origin and fate of the sperm transcripts remain subject to discussion.

متن کاملDevelopment of an Alu-PCR Amplified YAC Probe Suitable for Enumeration of Chromosome 13 on Uncultured Lymphocytes and Amniocytes by Fluorescence in situ Hybridization
The main objective of the present study was to develop an efficient and reliable probe to be routinely used for detection of chromosome 13 copy numbers by interphase FISH. To achieve this, a Yeast Artificial Chromosome (YAC) containing sequences specific for human 13q12 (744D11), was cultured and the whole yeast genomic DNA was extracted. The human insert within the isolated DNA was amplified b...
متن کاملP-1: Leptin mRNA Is Present in Bovine EpididymalSpermatozoa
Background: The present study aimed to investigate the presence of leptin mRNA transcript in epididymal spermatozoa of Holstein cattle by RT-PCR analysis. Materials and Methods: To collect sperm from epididymis, the whole testes of three adult Holstein bulls were quickly removed after slaughtering. Samples were transferred to the laboratory on ice and subsequently the epididymal contents were c...
متن کاملRNA IN SPERM: TOWARDS AN IDENTIFICATION OF RNAs TRANSMITTED FROM SPERM TO ZYGOTE ON FERTILIZATION
INTRODUCTION It is widely recognized that all that fathers essentially contribute to the next generation is half of their genome, and mature sperm cells are transcriptionally and translationally inactive. However, an increasing number of reports have shown that RNAs were present in human sperm detected by using RT-PCR, differential display, in situ hybridization and microarrays [1]. One study s...
متن کاملDifferential display identifies overexpression of the USP36 gene, encoding a deubiquitinating enzyme, in ovarian cancer
OBJECTIVES To find potential diagnostic markers or therapeutic targets, we used differential display technique to identify genes that are over or under expressed in human ovarian cancer. METHODS Genes were initially identified by differential display between two human ovarian surface epithelium cultures and two ovarian cancer cell lines, A2780 and Caov-3. Genes were validated by relative quan...
